Objective

To conduct a structured assessment of disease-modifying therapy (DMT) targets, mechanisms, and current clinical development programs , based on published scientific and clinical evidence, to guide future research and development priorities in Type 1 Diabetes (T1D).

Background Rationale

Autoimmune diseases remain a significant unmet medical need, driving continued development of disease-modifying therapies (DMTs) aimed at modulating immune-mediated tissue damage and altering disease progression. A range of immunomodulatory approaches are being investigated across clinical stages, reflecting the growing diversity of therapeutic strategies in this field.

The autoimmune therapeutic landscape continues to evolve, with increasing complexity across targets, mechanisms, and clinical development pathways.

Accordingly, there is a need for a structured, evidence-based assessment of DMT targets, mechanisms, and publicly reported clinical development programs based on published scientific and clinical data, to inform understanding of the current landscape and guide future research and development priorities.

Description of Project

Multiple immunotherapies have demonstrated encouraging Phase 2 efficacy signals in Stage 2 and Stage 3, highlighting the potential of immune-modulating approaches to alter disease progression. However, relatively few programs have advanced to pivotal (Phase 3) trials, reflecting scientific, clinical, and regulatory complexities, as well as the challenges associated with evaluating an increasingly diverse therapeutic landscape.

In this evolving field, there is a growing need for a structured, evidence-based review of disease-modifying therapy (DMT) targets, mechanisms, and publicly available development programs. Based on published and publicly disclosed scientific and clinical information, such an effort could provide a comprehensive and current perspective on the field and support strategic evaluation of emerging opportunities.

Anticipated Outcome

This study is expected to provide a structured, evidence-based overview of disease-modifying therapy targets, mechanisms, and publicly reported clinical development programs across autoimmune diseases, highlighting key trends, translational challenges, and areas of active therapeutic exploration to support informed research and development prioritization within T1D.

Relevance to T1D

Type 1 Diabetes represents a prototypical autoimmune disease in which disease-modifying approaches targeting immune pathways are actively being investigated. Insights from the broader autoimmune therapeutic landscape may therefore help providing mechanistic strategies, clinical development trends, and translational challenges relevant to T1D disease modification.
